Abstract

A Drug-Drug Interaction (DDI) is defined as an association between two drugs where the situation in which one drug affect the activity of another when they both administrated together.Effect of DDI are of two kind, one is Positive DDI and another is Undesirable DDI.Positive DDIs can usually improve the therapeutic effects of patients and reduce suffering of patients but undesirable DDIs cause the major cause of adverse drug reactions and result in the drug withdrawal from the market and the patient death.Study about DDI’s are of three kind and they are pharmacodynamic,pharmacokinetic,pharmaceutic.Actually multi drug treatment is done to reduce patient suffering,improve treatment effect and increase overall survival rate.Therefore, identifying DDIs has become a key component of the drug development and disease treatment.There exists traditional and computational method for discovering DDI’s.Traditional methods are named as In silico,In vivo,In vitro methods .In this study, a novel method to predict DDIs based on the Integrated Similarity and Semi-Supervised Learning (DDI-IS-SL) is proposed.DDI-IS-SL integrates the drug chemical, biological and phenotype data to calculate the feature similarity of drugs with the cosine similarity method. The Gaussian Interaction Profile kernel similarity of drugs is also calculated based on known DDIs. Next step is combining two results to get final similarity.Then a semi-supervised learning method Regularized Least Squares classifier(RLS) is used to calculate the interaction possibility scores of drug-drug pairs.Evaluation metrics used in this model are 5 fold cross validation,10 fold cross validation and denovo drug validation and advantages of this model when we compare with other model are better prediction performance and need only shorter computation time.
